Charlotte is really fortunate to have a 500'+ Tower under Construction as not many other US Cities do currently.   Seems like there a lot of talk about Companies reducing the need for Office Space and that will be reflected in Developers building less SF.

Went out for a random top-down drive late last night and made our way back home down Tryon St. and this site was humming at 1045P.  I didn't realize they are working on this... 24hrs/day?

Yes - It's Typical for Major Towers (500'+) to have 24 hr activity.  Usually big concrete pours at night and Formwork Stripping & Assembly during the Day Shift.  Probably true for some smaller Towers too, just depends on the Contractor if their able to coordinate such undertakings.

From that one college street angled view of the corner my count has them working on the 24th floor perimeter and floor system. So I think they have 16 -18 more floors to go! Plus or minus one depending on wether or not they actually label the 13th floor the 13th floor.
What was the final count for this building supposed to be again 40 or 42?

I don't remember. I remember the height was reduced not not sure about the floor count.

I can confirm there is no 13th floor labeled since I have a PDF drawing related to the work I'll be doing.  It skips from 12 to 14 and 41 is the last floor.
